Depends what your applying it to. If balsa it will raise the grain so yes sand lightly. It is probably a good idea to sand anyway. How many coats? - what are you trying to achieve and what are you sealing? Again, balsa will require quite  a few coats as it will soak up the sealer. If you are looking to fill the grain in ply, then again keep going until you get the surface you are looking for. In this case sand more heavily between coats so you are filling the grain without adding too much depth on the flat bits.
